Abstract

v(D)J recombination: for the community of immunologists and developmental biologists, the molecular route by which B and T lymphocytes acquire their unique function of affording adaptive immunity. Yet, for many-from experienced scientists to trainees-it represents a (rather too) sophisticated process whose true insight is excessively demanding. However, when not simplyconsidered as a private ground for a few aficionados, it can be seen as a way of understanding how maturelympho­ cytes carry on their basic functions. For the group of aficionados-which includes this editor-it is an elegant paradigm featuring many fascinating evolutionary achievements of which the biological world alone has the secret. These include a subtle biochemical principle most likelyhijacked some 470 million years ago from an ancestral gene invader and since then cleverly adapted by jawed vertebrates to precisely cleave and rearrange their antigen receptor (Ig andTCR)loci. This invader would itself have assigned the services of the nonhomologous end joining (NHEJ) DNArepair machinery as well as various DNApolymerases or transferases to work in concert with developmental clues in lymphoid cell lineages to generate an immune repertoire and efficient host surveillance while avoiding autoimmunity. Recently, important new refinements in these systems have emerged, continuing to challenge ourknowledge andbeliefs. Abstract

Chemokine Receptors in Cancer summarizes the growing body of evidence that several chemokine receptors contribute to tumor behavior. Chemokine receptors were first identified on leukocytes and mediate directed migration of many host cells to sites of ligand expression. It is now well established that most malignant cells also express one or more chemokine receptor. This book describes our current understanding regarding how chemokine receptors contribute to tumor cell migration as well as cell survival and proliferation. The function of chemokine receptors expressed on host cells including antitumor immune effector cells as well as angiostatic and angiogeneic functions of chemokines acting on endothelial cells are described. Abstract

There is a strong interest in transthyretin (TTR) in connection with protein evolution, medical and clinical research. Thus, this is an exciting time for experts in TTR research to come together to write a monograph covering both the basic and the clinical research into TTR. Transthyretin is a protein found in human blood and cerebrospinal fluid. It is directly involved in the transport of thyroid hormones, and indirectly in that of retinol. These hormones are essential for normal growth and development, particularly that of the brain. Spontaneous and inherited diseases affecting transthyretin result in amyloidosis. More than 80 point mutations in transthyretin lead to a variety of illnesses. Liver transplants are the most common treatment, although much research is also being carried out in drug therapies. The evolution of transthyretin has been extensively investigated – from humans to bacteria, invertebrate animals and plants. The structure of the protein has not changed, but its function has changed significantly. This is a most exciting example for the study of the evolution of protein structure-function relationships. This monograph will bring the reader up to date on the latest developments and discoveries.

Abstract

One chapter focuses on regulation of the blood brain barrier function by various neuropeptides, proteins, receptors, and transporters. Another is concerned with the modulation of higher brain functions by neuropeptides and biogenic monoamines. Yet another chapter presents research on ischemic neuronal damage and hippocampal neurogenesis in the adult mouse. Morphological or physiological techniques to study neuropeptides and neuromodulators influencing higher-order or brain-stem functions are given particular attention. The use of bio-imaging tools such as brain navigation systems and fMRIs with patients in a clinical setting creates new possibilities for investigation of human brain function and specialization of treatment.
